چکیده

Brillouin microscopy has recently emerged as powerful technique to characterize the mechanical properties of biological tissue, cell and biomaterials. However, the potential of Brillouin microscopy is currently limited to transparent samples, because Brillouin spectrometers do not have sufficient spectral extinction to reject the predominant non-Brillouin scattered light of turbid media. To overcome this issue, we developed a spectrometer composed of a two VIPA stages and a multi-pass Fabry-Perot interferometer. The Fabry-Perot etalon acts as an ultranarrow band-pass filter for Brillouin light with high spectral extinction and low loss. We report background-free Brillouin spectra from Intralipid solutions and up to 100 microns deep within chicken muscle tissue.

Multistage VIPA etalons for high-extinction parallel Brillouin spectroscopy

We demonstrate a high-resolution high-extinction parallel spectrometer for Brillouin spectroscopy of turbid samples. Cascading multiple VIPA etalons in the cross-axis configuration allowed us to achieve a high extinction ratio of up to 80 dB with sub-GHz resolution. Using a three-stage VIPA, we obtained the Brillouin spectra from Intralipid solutions at concentrations up to 10%.

Spatially resolved Brillouin spectroscopy to determine the rheological properties of the eye lens

Presbyopia is closely associated with the loss of accommodation, and hence with a decline in the viscoelastic properties of the human eye lens. In this article we describe a method for obtaining spatially resolved in vivo measurements of the rheological properties of the eye lens, based on the spectroscopic analysis of spontaneous Brillouin scattering using a virtually imaged phased array (VIPA...

The study of propagation of a femtosecond laser pulse in the breast tissue

In this paper, the evaluation of time profile of a femtosecond pulse laser propagated through biological tissues is studied. The majority of the biological tissues with a high scattering anisotropy must be considered as turbid media, that their optical responses are complicated. To study the propagation of ultra-short pulse in turbid media, the diffuse equation is used. In this study, the analy...

High Speed Sub-GHz Spectrometer for Brillouin Scattering Analysis.

The goal of this protocol is to build a parallel high-extinction and high-resolution optical Brillouin spectrometer. Brillouin spectroscopy is a non-contact measurement method that can be used to obtain direct readouts of viscoelastic material properties. It has been a useful tool in material characterization, structural monitoring and environmental sensing. In the past, Brillouin spectroscopy ...
